UBB events often produce large deformations of the floor and subsequent acceleration of the lower limb that are not sufficiently mitigated by the combat boot, leaving the calcaneus bone vulnerable to injury. Biomechanical experiments simulating UBB loading scenarios were conducted in a laboratory environment using isolated postmortem human subject (PMHS) leg components. Each leg component was tested twice one sub-injurious test followed by a injury-targeted test. This enabled the use of interval censoring for each specimen in the survival statistical analysis to generate the human injury probability curves (HIPCs). Foot contact forces were measured in both the hindfoot and forefoot. Strains and acoustic emission signals at the calcaneus and distal tibia were utilized to determine injury timing. The footplate velocities of the injury tests ranged 8-13 m/s with time-to-peak velocity of 1.8-2.5 ms while the velocities of non-injury tests ranged from 4 to 6 m/s with the same time-to-peak. The majority of the injuries were severe calcaneus fractures (Sanders III-IV). Secondary injuries included fractures to the distal tibia, talus, cuboid and cuneiform. These injury outcomes were found to be consistent with those reported in UBB injury literature. The HIPCs for the severe calcaneus fracture were developed using the vertical heel contact force as the injury correlation measure through survival analysis statistical method in the form of lognormal function. This work represents the first set of HIPCs dedicated to the severe calcaneus fracture using the biomechanical force measurement closest to the injury location. This injury probability curve will enable biomechanical response validation of computational models, development of ATD injury assessment reference curve, and injury prediction capability for computational models or ATDs in the UBB environment.Mechanical forces are essential for proper growth and remodeling of the primitive pharyngeal arch arteries (PAAs) into the great vessels of the heart. Despite general acknowledgement of a hemodynamic-malformation link, the direct correlation between hemodynamics and PAA morphogenesis remains poorly understood. The elusiveness is largely due to difficulty in performing isolated hemodynamic perturbations and quantifying changes in-vivo. Previous in-vivo arch artery occlusion/ablation experiments either did not isolate the effects of hemodynamics, did not analyze the results in a 3D context or did not consider the effects of varying degrees of occlusion. Here, we overcome these limitations by combining minimally invasive occlusion experiments in the avian embryo with 3D anatomical models of development and in-silico testing of experimental phenomenon. We detail morphological and hemodynamic changes 24 hours post vessel occlusion. PT2385 3D anatomical models showed that occlusion geometries had more circular cross-sectional areas and more elongated arches than their control counterparts. Computational fluid dynamics revealed a marked change in wall shear stress-morphology trends. Instantaneous (in-silico) occlusion models provided mechanistic insights into the dynamic vessel adaptation process, predicting pressure-area trends for a number of experimental occlusion arches. Individuals may also be at risk for substance use offending and substance use problems based on genetic predisposition. We examined a mediation model in which polygenic risk for aggression predicted adult substance use disorder diagnoses (SUD) via substance use offending in emerging adulthood. In addition, we explored for potential attenuation of genetic influences on these outcomes by a family-based intervention, the Family Check-Up (FCU). Secondary data analyses based upon the Project Alliance 1 sample was conducted among those with genetic data (n = 631; 322 from control and 309 from FCU intervention). The sample was ethnically diverse (30% African American, 44% European American, 6% Latinx, 4% Asian American, 3% Native American, and 13% Other). Greater polygenic risk for aggression was found to increase risk for substance use violations (age 19-23), which in turn was associated with greater likelihood of being diagnosed with SUD at age 27. A gene-by-intervention effect was found in which individuals in the control group had greater risk for SUD with increasing polygenic risk for aggression. Some convergence in results was found when replicating analyses in African American and European American subgroups. Results imply that genetic predisposition may increase risk for problematic substance use later in life via antisocial behavior, such as substance use offending, and that this can be attenuated by a family-centered intervention.
