Within HPV-positive tumors, there is wide morphologic diversity with numerous histologic subtypes of SCC. There are also variable degrees of keratinization, anaplasia, stromal fibrosis, and maturing squamous differentiation. Unlike in the uterine cervix, where associations between HPV types and lineages/sublineages within types have been investigated with some clear correlations identified, little to no data exists for oropharyngeal SCC. In this study, for a large cohort of oropharyngeal SCC patients, we performed RTPCR for high-risk HPV. https://www.selleckchem.com/products/gw788388.html For the HPV positive patients, we sequenced the DNA of the entire HPV16 genome and determined lineages and sublineages, correlating HPV status, genotype, and HPV16 lineages/sublineages with SCC subtype and various histologic features. Of the 259 patients, 224 (86.5%) were high-risk HPV positive, of which 210/224 (93.8%) were HPV type 16 and 6/224 (2.7%) HPV type 33. Of the four HPV16 lineages, A was the most frequent (192/214 or 89.8%) and of the HPV16 A sublineages, A1 was the most frequent (112/210 or 53.3%). Patients with HPV negative tumors were more often keratinizing vs other types (23/35 or 65.7%) and thus more likely to have more maturing squamous differentiation and stromal desmoplasia. There was no significant correlation between HPV type (16 versus other), between HPV16 lineage (A versus others), or HPV16 A sublineages (A1 or A2 versus others) and morphologic type of SCC nor the various morphologic features of anaplasia/multinucleation, degree of keratinization, nor amount of stromal desmoplasia. In summary, in our cohort, there was no correlation between the type of HPV, the HPV 16 lineage or sublineage, and any of the histologic features or morphologic SCC subtypes.Oral squamous cell carcinoma (OSCC) occasionally occurs in young patients and is likely to be distinct from OSCC in older patients. In this retrospective study, we described the clinicopathologic features and outcome of 150 OSCCs that were diagnosed in patients 40-year-old or younger. Most patients (63%) were non-smokers. The most common site of the primary tumor was oral tongue (n = 131, 87%), followed by gingiva (n = 9), buccal mucosa (n = 8) and lip (n = 2). The median patients’ age at presentation was 34 (range 16-40). Seven patients (5%) had Fanconi anemia with the gingiva being the most common location (4/7, 57%). All OSCCs were of keratinizing type. All cases tested for high-risk HPV (n = 34) were negative. On univariate analysis, high tumor budding was associated with decreased overall survival (OS) and distant metastasis free survival (DMFS), pattern of invasion correlated with OS and tumors with high stromal tumor infiltrating lymphocytes (sTIL) were associated with improved locoregional recurrence free survival (LRFS). Compared with patients 31 to 40-year-old, OSCC in the younger group was associated with significant less alcohol consumption (p = 0.011) and decreased DSS (p = 0.003) and DMFS (p = 0.023). On multivariate analysis, younger age (30 years or younger) was an independent prognostic factor for worse OS and DSS, whereas histologic grade was an independent prognostic factor for DSS. In summary, most OSCC in young patients occurred in non-smokers and did not occur in association with Fanconi anemia. Independent prognostic factors included age at presentation (30 years or younger) for OS and DSS, and histologic grade for DSS.The velopharyngeal valve regulates the opening between the nasal and oral cavities. The lack of complete closure is especially problematic in speech because inappropriate leakage of airflow and/or sound into the nasal cavity causes abnormal sound production and increased nasality. The purpose of this study is to use the large eddy simulation approach to examine changes in sound source mechanisms as the size of the opening changes during the production of a sibilant sound. The baseline geometry of the model is based on the pharyngeal airway of a subject having a small velopharyngeal opening while sustaining a sibilant sound. Modifications to the model are done by systematically widening or narrowing the opening (all else being equal). Results show that acoustic energy in the nasal cavity is directly related to the size of the velopharyngeal opening and that there is a critical size where the magnitude of Lighthill’s acoustics source in the nasal cavity is maximized. The far-field acoustic energy and its correlation with the sound source mechanisms are also dependent on the size of the velopharyngeal opening. Cervical cancer is one of the most common and fatal malignancies among females, and biomarkers are essential for assisting in its management. Kinesin family member 2A (KIF2A) has been exhibited to be a potential maker in various cancers; however, its role in cervical cancer has yet to be reported. Therefore, we aimed to assess the expression of KIF2A and its correlation with clinicopathological characteristics as well as survival profile in cervical cancer patients.
A hundred and thirty-five cervical cancer patients who underwent simple trachelectomy or radical hysterectomy were retrospectively analyzed. Tumor tissues and paired adjacent tissues were acquired, in which KIF2A mRNA and protein expressions were determined by reverse transcription quantitative polymerase chain reaction and immunohistochemistry assay, respectively. Disease-free survival (DFS) and overall survival (OS) were documented with a median follow-up duration of 28.0months.
KIF2A protein (P < 0.001) and mRNA (P < 0.001) expressions were both upregulated in tumor tissues compared to paired adjacent tissues in cervical cancer patients.
