    The purpose of this article is to review and discuss the factors affecting high pressure processing (HPP) in juices and beverages. The inactivation of microorganisms by HPP depends on numerous factors, including the magnitude of the pressure and the holding time, process temperature, compression and decompression rates, the microbiota, and the intrinsic properties of juices and beverages. Although extensive HPP research has been performed to characterize many of these factors, a number of issues, such as the rates of compression and decompression, still remain unresolved and need further investigation. In addition, some published results are conflicting and do not provide enough evidence to develop juice HPP “safe-harbor” parameters to achieve a minimum 5-log reduction of the pertinent microorganism and produce safe fruit juices and beverages.

    The aim of the study was to determine the levels of aflatoxin B1 (AFB1) and ochratoxin A (OTA) in durum wheat samples collected over six periods of time from two official warehouses (A and B) in Lebanon. The concentrations of AFB1 in wheat samples taken from both warehouses ranged from 1.05 to 7.36 μg/kg. Results showed that 23.3 and 25.3% of samples taken from warehouse A and warehouse B, respectively, had AFB1 levels >2 μg/kg. selleck compound For OTA, the contamination level of wheat samples taken from warehouse A and warehouse B was 52.0 and 44.6%, respectively, with concentrations ranging from 0.51 to 9.71 μg/kg.     This study optimized the method for ozone (O3) degradation of prometryn in the clam Ruditapes philippinarum and evaluated toxicity changes during ozone degradation. The gas chromatography method for the detection of prometryn was appropriately improved. The linear range was 5 to 500 ng/mL, with a correlation coefficient of 0.9964. The addition concentration of prometryn was 0.025 to 0.100 mg/kg, the recovery was 77.97 to 85.00%, the relative standard deviation (n = 6) was 2.36 to 7.86%, and the limit of detection was 0.3 μg/kg. Using the central composite design in two experiments, ozone as gas and ozone dissolved in water, the effect of degradation rate was studied on three variables ozone concentration, temperature, and exposure time. Ozone as gas and ozone dissolved in water have the same degradation effect on prometryn. The O3 concentration was 4.2 mg/L, the temperature was 40°C, the exposure time was 10 min, and the maximum degradation rate was 89.94 and 89.69% for the two experiments, respectively. In addition, the toxicity of ozone degradation products was evaluated using buffalo rat liver cells. After ozone treatment for 30 min, the toxicity of the ozone degradation products was reduced to 52.15% compared with that of prometryn itself. The toxicity of the ozone degradation products increased slightly when the ozonation time was prolonged; the toxicity at 180 min was greater than that of the parent compound prometryn. Overall, the application of ozone degradation of pesticide residues is a promising new technology. This study determined better degradation conditions for prometryn in R. philippinarum and also provided a theoretical basis for the widespread use of ozone technology in the future.

    Hospital-acquired infections caused by extended-spectrum β-lactamase (ESBL)-producing Escherichia coli are a global problem. Healthy people can carry ESBL-producing E. coli in the intestines; thus, E. coli from healthy people can potentially cause hospital-acquired infections. Therefore, the transmission routes of ESBL-producing E. coli from healthy persons should be determined. A foodborne outbreak of human norovirus (HuNoV) GII occurred at a restaurant in Shizuoka, Japan, in 2018. E. coli O25H4 was isolated from some of the HuNoV-infected customers. Pulsed-field gel electrophoresis showed that these E. coli O25H4 strains originated from one clone. Because the only epidemiological link among the customers was eating food from this restaurant, the customers were concurrently infected with E. coli O25H4 and HuNoV GII via the restaurant food. Whole genome analysis revealed that the E. coli O25H4 strains possessed genes for regulating intracellular iron and expressing the flagellum and flagella. Extraintestinal pathogenic E. coli often express these genes on the chromosome. Additionally, the E. coli O25H4 strains had plasmids harboring nine antimicrobial resistance genes. These strains harbored ESBL-encoding blaCTX-M-14 genes on two loci of the chromosome and had higher ESBL activity. Multilocus sequence typing and fimH subtyping revealed that the E. coli O25H4 strains from the outbreak belonged to the subclonal group, ST131-fimH30R, which has been driving ESBL epidemics in Japan. Because the E. coli O25H4 strains isolated in the outbreak belonged to a subclonal group spreading in Japan, foods contaminated with ESBL-producing E. coli might contribute to spreading these strains among healthy persons. The isolated E. coli O25H4 strains produced ESBL and contained plasmids with multiple antimicrobial resistance genes, which may make it difficult to select antimicrobials for treating extraintestinal infections caused by these strains.
